HLA Cross-match Tests

About the test

This test determines if the patient has antibodies to a particular donor. Different cross-match techniques have been designed to identify pathogenic antibodies, relevant to transplantation.

Turn Around Time:
Routine: 3 Working days
Emergency: 6 to 8 Hours

CCDC

This is a comprehensive classic complement dependent cytotoxicity Crossmatch (CDCXM).
It comprises of the following:

  • Allo & Auto- T cell & B cell crossmatch

  • The T cell crossmatch with AHG enhanced to increase the sensitivity of the crossmatch

  • A DTT treatment will be performed if required

  • The Allo crossmatches will run in dilutions

BCDC

This is a basic CDCXM.
It comprises of the following:

  • Allo and Auto crossmatch (XM) with total lymphocytes

  • AHG enhancement is added to increase the sensitivity of the crossmatch

  • The test is run in dilutions

FCXM

This is a flow cytometry based crossmatch. It is 3-4 times more sensitive than the comprehensive CDCXM.
It comprises of the following:

  • Allo & Auto- T cell & B cell crossmatch

  • The Allo test is run in dilutions to overcome prozone effect

AFCXM

This is a flow cytometry based crossmatch. It is 3-4 times more sensitive than the comprehensive CDCXM.
It comprises of the following:

  • Allo T cell & B cell crossmatch

  • The test is run in dilutions to overcome prozone effect

PFCXM

Pronase enhancement will be offered if false positive B cell XM is noted with a history of Rituxumab.

Antibody Titration for positive XM

All the XMs as described above in each category with added dilutions till XM turns negative

All the XMs as described above in each category with added dilutions till XM turns negative

DCCDC

All the XMs as described above in each category with added dilutions till XM turns negative

DFCXM

All the XMs as described above in each category with added dilutions till XM turns negative

DPFCXM

All the XMs as described above in each category with added dilutions till XM turns negative

HLA XM Trend analysis

All the XMs as described above in each category are done with current, peak, historic sera (this test can be done only if serum samples of patients are available)
